چکیده

Blood–Brain Barrier In article number 2100812 by Smriti Singh, Agnieszka Morgenroth, and co-workers, Matrix metalloprotease (MMP) sensitive nanogels are synthesized for effective delivery of a radiopharmaceutical ([125I]ITdU) across the blood-brain barrier (BBB). The show efficient transcytosis in vitro model BBB co-cultured with glioblastoma cells. Once transcytosed, degrade an up-regulated MMP2/9 environment release radiopharmaceutical, which gets incorporated into DNA tumor cells thymidine salvage pathway.

Dual responsive supramolecular nanogels for intracellular drug delivery.

Supramolecular nanogels cross-linked by host-guest interaction between dextran grafted benzimidazole (Dex-g-BM) and thiol-β-cyclodextrin were designed. Their special supramolecular pH-sensitivity under acidic conditions (pH < 6, within the range of malignant cellular endosomes) and reduction sensitivity in response to biologically relevant stimuli will be of great advantage to the future of can...

Targeting blood-brain-barrier transcytosis – perspectives for drug delivery

Efficient transcytosis across the blood-brain-barrier (BBB) is an important strategy for accessing drug targets within the central nervous system (CNS). Despite extensive research the number of studies reporting successful delivery of macromolecules or macromolecular complexes to the CNS has remained very low. In order to expand current research it is important to know which receptors are selec...

Smuggling Drugs into the Brain: An Overview of Ligands Targeting Transcytosis for Drug Delivery across the Blood–Brain Barrier

The blood-brain barrier acts as a physical barrier that prevents free entry of blood-derived substances, including those intended for therapeutic applications. The development of molecular Trojan horses is a promising drug targeting technology that allows for non-invasive delivery of therapeutics into the brain. This concept relies on the application of natural or genetically engineered protein...

Nanogels for intracellular delivery of biotherapeutics.

Many biomolecules, such as proteins and genes, are presently used as therapeutics. However, their delivery to target sites inside cells is challenging because of their large molecular size, difficulties to pass cellular membranes and their susceptibility for enzymatic and chemical degradation. Nanogels, three-dimensional networks of hydrophilic polymers, are attractive carrier systems for these...

Stimulus-Responsive Short Peptide Nanogels for Controlled Intracellular Drug Release and for Overcoming Tumor Resistance.

Multidrug resistance (MDR) poses a major burden to cancer treatment. As one important factor contributing to MDR, overexpression of P-glycoprotein (P-gp) results in a reduced intracellular drug accumulation. Hence, the ability to effectively block the efflux protein and to accumulate the therapeutics in cancer cells is of great significance in clinical practice. In this work, we successfully de...
